The cathedral is constructed primarily of stone and features a neoclassical architectural style. Its facade is characterized by two bell towers flanking a central pediment. The exterior walls are painted a pale yellow, with white trim around windows and doorways. The main dome is octagonal and covered in terracotta tiles.

The cathedral is situated in Ignacio Agramonte Plaza, the main square of Camagüey. It is approximately 1km from the Camagüey train station. Vehicle access to the plaza is restricted during certain hours, but pedestrian access is open. Limited street parking is available on surrounding avenues, with larger parking lots located about 500 meters away. Bus lines 1 and 3 stop within a 200-meter radius of the plaza.

The current structure was built between 1747 and 1850, replacing an earlier church. The original construction was initiated by Bishop Francisco de la Lastra y Fuertes. Its purpose was to serve as the primary religious center for the growing city of Santa María del Puerto del Príncipe, later renamed Camagüey.
