Marsaxlokk Bay is situated approximately 10 kilometers southeast of Valletta, Malta's capital. Access by car is via Triq il-Qajjenza or Triq Delimara. Public parking is available along the waterfront and in designated lots near the village center. Public transport is serviced by Malta Public Transport bus routes, including route 81 from Valletta and route 119 from Malta International Airport.

Marsaxlokk Bay is a natural geological indentation, historically utilized as a harbor since Phoenician times. The village developed around this natural port, becoming a primary fishing hub. Its strategic location made it a landing point for Ottoman forces during the Great Siege of Malta in 1565 and for Napoleon's invasion in 1798.

The eyes painted on the bow of every Luzzu boat are known as the "Eye of Osiris" or "Eye of Horus." This ancient Phoenician custom is believed to protect the fishermen from evil spirits and ensure a safe return from the sea.
