Melrose is a two-story Greek Revival mansion constructed primarily of brick, stuccoed and painted white. The main structure features a prominent central portico with four Doric columns supporting a pediment. The roof is low-pitched and hipped. Melrose is situated approximately 5 miles (8 km) east of downtown Natchez, Mississippi. Access is via U.S. Route 84 East. The estate has a dedicated parking area for visitors. No public transportation directly serves the site.

Construction of Melrose began in 1841 and was completed in 1845. It was built by John Henderson, a wealthy planter. The original purpose was as a residence and center for a large cotton plantation. The estate's design reflects the prosperity of the cotton era in the Mississippi Delta.
