The park is situated approximately 100 kilometers north of Manta. Access to the park's terrestrial sector is via the E15 coastal highway. The main entrance is near Puerto Cayo. Public transport options include interprovincial buses traveling along the E15, with stops at Puerto Cayo. Parking is available at designated areas near the park entrance and visitor centers. Access to Isla de la Plata requires boat transfer from Puerto López, which is located 5 kilometers south of the park's main terrestrial entrance.

Parque Nacional Machalilla was established on July 18, 1979. Its creation aimed to protect the unique dry forest ecosystem and the marine life of the Ecuadorian coast. Ecologically, the park represents a transition zone between tropical dry forest and marine environments, supporting a high level of biodiversity.
